Effects of PTHrP on AKT, ERK, and JNK signaling pathways in hDPCs. (a) Effects of PTHrP on AKT, ERK, and JNK phosphorylation were assessed by Western blot analysis. Phosphorylation levels of AKT, ERK, and JNK were increased by PTHrP. (b–e) Effects of PTHrP and LY294002, U0126, and SP600125 on ALP staining and alizarin red S staining. The cells were pretreated with or without 10 μM LY294002, 10 μM U0126, or 10 μM SP600125 for one hour after treatment with 1 nM PTHrP. ALP staining (b, c) and alizarin red S staining (d, e) are shown. LY294002, U1026, and SP600125 significantly decreased PTHrP-induced ALP staining and mineralization (***P < 0.001 compared with the group treated with 1 nM PTHrP)
